A Spectrum Sensing Method based on Gold Nanorods for the Detection of Hepatitis B Virus and Human Papilloma Virus

In this study, we presented a novel extinction spectrum biosensing method based on gold nanorods (AuNRs) using a hyperspectral unmixing algorithm for the detection of hepatitis B virus (HBV) and human papillomavirus (HPV). A sandwich complex of MB-target DNA-AuNRs was formed by a two-step hybridization process using two sets of complementary sequences of the target DNA. After cleaning, dissociation, and aggregation, we obtained MB-free AuNRs. Almost every AuNR in the solution originated from a single HPV or HBV fragment. As a result, the focus shifted from detecting HPV/HBV fragments to detecting AuNRs. We demonstrated the method's feasibility by performing multi-concentration experiments on the HBV fragments, and the results of the combined HBV and HPV detection also confirmed the potential of the technique for multiplex analysis. Our method is rapid and does not require complex ex-perimental procedures and costly instrumentation. These results indicate that the proposed sensing method can be ap-plied to practical biomedical applications.
