Abstract:
By modern micromachining technique and volume wafer fabrication we produced a single-use patch-clamp chip which is contacted via a silicon rubber packaging. The unique microfluidic concept facilitates high-quality whole-cell recordings with stable gigaseals, rapid compound application and washout necessary for investigations on compound effects on both voltage and ligand gated ion channels in secondary screening and safety pharmacology. We successfully used the chip in the CytoPatchTM device for drug testing on CHO cells expressing hERG channels.
